Brief summary of problem (put in title as well):
Trying to transmit using the PTT button does not put the IC-7300 into Xmit mode.
Radio Model: IC-7300
Connectivity (USB/Ethernet/Wifi/Other): USB
Operating System: Windows 10
wfview version (press “About”): 2.03
Checked the wfview manual (Y/N): Y many times
Checked the wfview FAQ (Y/N): Y
Tried to google it (Y/N/NA): Y
What I’ve tried:
A terminal emulator program does toggle PTT when the DTR is enabled,
Another program, HRD does toggle PTT when the transmit button is pushed, (so serial interface is working),
Using Windows 10, unfortunately,
Settings: Radio Access → PTT Type is set to DTR,
Settings: User Interface → Enable PTT Controls is checked. Also tried unchecked,
Settings: Serial (USB) is checked,
All other functions seem to be working beautifully. (Very impressed with this)
Can’t think of anything else to try.
I’m very impressed with the program. Clean interface, easy to use, understandable and well thought out help files and user manual. Now if only I can get the PTT to work since when the program is connected the serial port cannot be shared with another program. Looking forward to some advise.
RTS will work but there is an issue with DTR which “should” work in the next version, as @eliggett said though, why are you using DTR when PTT over C-IV is available?

I assume the question why am I using USB instead of CI-V is rhetorical and I assume you mean USB-CI-V and not the external CI-V. But I am not aware the advantages that using USB-CIV instead of USB has, but I’ll Google it. Looks like I’m in store for an education.
I’m also trying com0com and haven’t been successful yet, but will continue trying.

We are talking about two different things, CI-V is both an interface and a protocol. The CI-V protocol is the commands that wfview uses to communicate with the rig, so there is no need to use DTR/RTS as the CI-V command to set PTT on/off is integrated within wfview.
The RTS/DTR for PTT option is ONLY supposed to be used with non-usb rigs like the IC718.
